摘要
目的 :检测Ca(OH) 2 激动体外培养的人牙髓细胞内Ca2 +的释放和钙释放通道。方法 :在RPMI16 40培养液中分别加入Ca(OH) 2 、Ca(OH) 2 和肝素、Ca(OH) 2 和普鲁卡因 ,培养第 6代人牙髓细胞 ,用Fluo - 3荧光探针负载后 ,再次用Ca(OH) 2 作用于牙髓细胞 ,用激光共聚焦显微镜检测胞内Ca2 +。结果 :含Ca(OH) 2 、Ca(OH) 2和普鲁卡因的培养液培养的细胞内Ca2 +浓度升高 ;含Ca(OH) 2 和肝素的培养液培养的细胞内Ca2 +浓度无变化。结论 :Ca(OH) 2 可激动人牙髓细胞内三磷酸肌醇受体 (IP3R) ,钙释放通道开放 ,使细胞内Ca2
AIM: To investigate intracellular calcium ion release by Ca(OH) 2 stimulus in human pulp cells in vitro and to detect a system of calcium channel. METHODS: The sixth generation of human pulp cells was cultured in RPMI-1640 medium with Ca(OH) 2, Ca(OH) 2 and heparin, Ca(OH) 2 and procaine. With the Fluo-3/AM Probe loaded, the Ca(OH) 2 was added to the medium and then the intracellular calcium level was detected by laser confocal microscope. RESULTS: Intracellular calcium concentration increased in pulp cells treated with Ca(OH) 2, Ca(OH) 2 and procaine, while it didn't change in with Ca(OH) 2 heparin treatment. CONCLUSION: Ca(OH) 2 can stimulate the intracellular calcium release channel of inositol triphosphate receptor open in pulp cells in vitro and increase the level of intracellular calcium concentration.
