粉葛种苗离体繁殖技术初步研究 被引量:10

摘要 为获得粉葛的离体繁殖技术,以地理标志火山粉葛嫩枝为外植体,通过调整基本培养基、生长调节剂种类和配比,选出适合的诱导、继代增殖和生根培养基配方。结果显示,粉葛嫩茎尖是诱导愈伤组织的最佳材料。培养基MS+6-BA 0.1 mg/L+2,4-D 0.1 mg/L有利于粉葛茎段侧芽的萌发,而MS+6-BA 0.5 mg/L+2,4-D 1.0 mg/L则能高效诱导茎段产生愈伤组织。但在对粉葛嫩茎尖的诱导试验中,培养基MS+6-BA 0.1 mg/L+2,4-D 0.5 mg/L体现出促进芽萌发、愈伤组织形成并高效分化丛生芽的综合特点。培养基MS+NAA 0.5 mg/L能较好地诱导粉葛试管苗形成不定根。 To provide much more high quality seeding for farmer,the different factors affect on buds and tissues of Huoshan Kudzu(Pueraria thomsonii Benth) in vitro.The tender branch of Kudzu,include shoots,stems,were used as explants and cultivated in different media.The optimum media for inducing buds,proliferation and rooting were selected by adjusting the kinds and doses of plant hormones and special compounds.The result showed that the tender stem apexes of Pueraria thomsonii Benth was the best materials for inducing tissues.The optimal media to induce sprouting from stems were MS + 6-BA 0.1 mg/L + 2,4-D 0.1 mg/L.And media MS + 6-BA 0.5 mg/L + 2,4-D 1.0 mg/L was suitable for induce much more tissues from stems.Otherwise,media MS + 6-BA 0.1 mg/L + 2,4-D 0.5 mg/ L had the abilities to induce sprouting,tissues formation and buds polarization effectually.The media MS + NAA 0.5 mg/L was used for roots inducing,the rooting rate was 73.3%.
