芪归银方对多重耐药铜绿假单胞菌感染大鼠淋巴细胞增殖的影响 被引量:23

Effect of Qiguiyin Formula on the Lymphocyte Proliferation in Rats with Multidrug-Resistant Pseudomonas Aeruginosa Infection

摘要 目的探讨芪归银方对多重耐药铜绿假单胞菌感染大鼠的免疫调节作用。方法将40只SD大鼠随机分为空白组、模型组、西药对照组、芪归银方组、中西医结合组,每组8只。除空白组外其余各组大鼠将多重耐药铜绿假单胞菌菌液同2.0%西黄芪胶1∶1混合后8ml/kg腹腔注射,注射后空白组和模型组给予蒸馏水2ml灌胃,西药对照组给予头孢他啶0.45g/kg肌肉注射,芪归银方组给予芪归银方提取物2ml(1.0g/ml)灌胃,中西医结合组给予芪归银方提取物灌胃和头孢他啶肌肉注射,各组腹腔感染后3h留取脾脏组织标本,运用MTT法检测各组大鼠T、B淋巴细胞的增殖水平。结果模型组大鼠脾脏中T、B淋巴细胞增殖水平较空白组升高(P<0.05或P<0.01);西药对照组大鼠脾脏中T、B淋巴细胞增殖水平较空白组降低(P<0.05或P<0.01);芪归银方组和中西医结合组大鼠脾脏中T、B淋巴细胞增殖水平较空白组差异无统计学意义(P>0.05);西药对照组、芪归银方组和中西医结合组大鼠脾脏中T、B淋巴细胞增殖水平均较模型组降低(P<0.05或P<0.01)。结论芪归银方具有适度下调多重耐药铜绿假单胞菌感染大鼠淋巴细胞增殖能力,与头孢他啶过度抑制免疫应答反应相比,其可使感染所导致的免疫紊乱趋于平衡。 Objective To study the immunomodulatory effect of Qiguiyin Formula [formula of Huangqi (Radix Astragali), Danggui (Angelica Sinensis) and Jinyinhua (Flos Lonicerae )1 on rats with multidrug-resistant pseudomonas aeruginosa infection. Methods The SD rats were randomized into the blank group, model group, western medicine control group, Qiguiyin Formula group and integrative medicine group, with 8 in each. The multi-drug resistant pseudomonas aeruginosa bacteria and 2.0% tragaeanth were mixed at one to one ratio and 8 ml/kg of the mixture was intraperitoneally injected to the rats in each group except the blank group. After injection, the blank group and model group were gavaged with 2 ml distilled water, the western medicine control group was intramuscularly injected with 0.45g/kg ceftazidime, the Qiguiyin Formula group was gavaged with 2 ml (1.0 g/ml) Qiguiyin Formula extract and the integrative medicine group was gavaged with Qiguiyin Formula extract and intramuscularly injected with ceftazidime. The spleen tissue specimens were taken 3 hours after intraperitoneal infection. The proliferation levels of T and B lymphocytes were detected by MTT method. Results Comparing with the blank group, the proliferation levels of T and B lymphocytes were significantly increased in the model group (P〈0.05 or P〈0.01). Comparing with the blank group, the proliferation levels of T and B lymphoeytes were significantly decreased in the western medicine control group (P〈0.05 or P〈0.01). Comparing with the blank group, the proliferation levels of T and B lymphoeytes were not significantly changed in the Qiguiyin Formula group and integrative medicine group (P 〉 0.05). Comparing with the model group, the proliferation levels of T and B lymphocytes were significantly decreased in the western medicine control group, Qiguiyin Formula group and integrative medicine group (P〈0.05 or P〈0.01). Conclusion Qiguiyin Formula can moderately down-regulate the lymphocyte proliferation in rats with multidrug-resistant pseudomonas aeruginosa infection. Comparing with the excessive inhibition of immune response by ceftazidime, Qiguiyin Formula can balance the immune disorders caused by infection.
