搜剔肺络方对肺纤维化大鼠转化生长因子β_1及Smad蛋白的影响 被引量:4

Effect of Souti Feiluo Formula on TGF-β_1,Smad3 and Smad 7 in Rats with Pulmonary Fibrosis

摘要 目的探讨搜剔肺络方对肺间质纤维化大鼠的作用及可能机制。方法 128只SD大鼠随机分为假手术组、模型组、中药组、西药组各32只;除假手术组外,其余各组一次性气管内滴注盐酸博莱霉素制备肺纤维化大鼠模型,假手术组气管内注射等体积生理盐水,造模后24h中药组给予搜剔肺络方0.68g/(kg·d)灌胃,西药组给予泼尼松龙5mg/(kg·d)灌胃,模型组和假手术组给予等体积的去离子水,每日1次,分别于第7、14、28、45天处死,取肺组织行病理学观察肺泡炎程度和肺纤维化程度,检测血清转化生长因子-β1(TGF-β1)含量及肺组织Smad3、Smad7蛋白的表达。结果与模型组比较,第7、14、45天中药组和西药组肺泡炎程度均明显减轻(P<0.05),第7、45天中药组和西药组肺纤维化程度明显减轻(P<0.05)。与模型组比较,中药组和西药组TGF-β1含量差异无统计学意义(P>0.05);各时间点中药组和西药组Smad3均较模型组明显减少(P<0.05),且中药组明显低于西药组(P<0.05);各时间点中药组和西药组Smad7较模型组均明显增加(P<0.05)。结论搜剔肺络方对博莱霉素致大鼠肺纤维化有一定的抑制作用,其机制可能与调节Smad信号表达有关。 Objective To study the mechanism of Souti Feiluo Formula (formula to treat pulmonary fibrosis) for pulmonary fibrosis. Methods Totally 128 rats were randomized into the sham operation group, model group, Chinese medication group and western medication group, with 32 in each. The pulmonary fibrosis models were established by intratracheal instillation of bleomycin in each group except the sham operation group. The sham operation group was given intratracheal instillation of normal saline. At the 24th hour after modeling, the Chinese medication group was gavaged with 0.68 g/(kg · d) Souti Feiluo Formula, the western medication group was gavaged with 5 mg/(kg ·d) prednisolone and the model group and sham operation group were gavaged with same amount of deionized water. The rats were sacrificed on the 7th, 14th, 28th and 45th day, respectively. The lung tissues were taken for pathological observation of the degree of pulmonary alveolitis and fibrosis. The content of transforming growth factor-β1(TGF-β1) and the expressions of Smad3 and Smad7 were detected. Results Comparing with the model group, the degree of alveolitis was significantly decreased on the 7th, 14th and 45th day (P〈0.05) and the degree of pulmonary fibrosis was significantly decreased on the 7th and 45th day (P〈0.05) in the Chinese medication group and western medication group. There was no significant difference between the model group, Chinese medication group and western medication groups in the content of TGF-β1 (P〉0.05). Comparing with the model group, the expression of Smad3 was significantly decreased in the Chinese medication and western medication groups (P〈0. 05) on each of point-in-time, especially in the Chinese medication group (P〈0. 05). Comparing with the model group, the expression of Smad7 was significantly increased in the Chinese medication and western medication groups on each of point-in-time (P〈0. 05) . Conclusion Souti Feiluo Formula can inhibit the pulmonary fibrosis caused by bleomyein and its mechanism may be relevant to regulating the expression of Smad.
