乳岩宁方联合依西美坦诱导绝经后裸鼠乳腺癌移植瘤细胞凋亡的实验研究

Study on Ruyanning plus Exemestane to induce apoptosis of MCF - 7 breast cancer on postmenopausal tumor - bearing nude mice

目的:探讨乳岩宁方药与依西美坦联合应用对乳腺癌的抑制作用及其作用机理。方法:采用双侧卵巢切除的方法制备绝经后裸鼠模型,将对数生长期的MCF-7细胞调整浓度为5×107/ml,每只接种于卵巢切除后的裸鼠右侧胸壁第二乳垫部皮下,0.2ml/只,待裸鼠长出肿瘤0.8 cm3后,将取得的瘤块切成1mm3大小,无菌条件下接种于裸小鼠右腋窝脂肪垫下,建立绝经后裸鼠移植瘤模型。选取荷瘤成功的裸鼠随机分为4组:对照组、依西美坦组、乳岩宁组、联合组(乳岩宁联合依西美坦)。记录各组裸鼠饮食、活动、皮毛色泽、死亡及体重变化情况;第21天,处死裸鼠取肿瘤,称重,计算抑瘤率;TUNEL法测肿瘤细胞凋亡。结果:体重:实验结束时,各用药组体重比较无显著差别,但是对比体重下降,联合组下降最少,为0.93±0.06g,依西美坦组下降最多,为1.61±0.12g,乳岩宁组为1.11±0.16g,各组间比较有统计学差异(P<0.05);瘤重:依西美坦组为0.197±0.021g、乳岩宁组为0.223±0.028g、联合组0.161±0.021g,肿瘤重量均低于对照组的0.298±0.022g,与对照组比较有非常显著差异(P﹤0.01);依西美坦组、乳岩宁组、联合组的抑瘤率分别为33.9%、25.2%、46.0%;TUNEL法检测裸鼠肿瘤组织细胞凋亡结果:凋亡指数(AI):联合组最高为(39.37±3.12)%,依西美坦组(31.41±2.89)%,乳岩宁组(27.89±2.94)%,与对照组相比有统计学差异(P<0.01);联合组与单独用药组比较差异均有统计学意义(P<0.01)。结论:乳岩宁对裸鼠MCF-7乳腺癌移植瘤有抑制作用,与依西美坦有协同作用,增加抑瘤率,其作用机制可能通过诱导肿瘤细胞凋亡来实现的。

Objective：To observe the influence of Ruyanning plus endocrine drug of Exemestane to inhibit the growth of tumor and induce apoptosis.Methods：Nude mice were injected MCF-7 cell at logarithmic phase subcutaneously.To cut the tumor blocks to 1 mm3 when tumor grew to 0.8 cm3,and transplanted it to other nude mice on the secondary breast pad of the right chest wall.40 mice were randomized into control group,Exemestane group,Ruyanning group and combination group （Ruyanning plus Exemestane） after MCF-7 tumor-bearing successfully.The change of diet,activity,death and weight was recorded.At the end of the experiment mice were killed,tumor was taken and weighed,the inhabitation of tumor was calculated.The change of pathomorphology was observed by HE staining.The apoptosis of tumor was detected by TUNEL.Results：Weight：Except control group,the decline of weight in combination group was the least,while in Exemestane group was the most,there was significant difference （P ＜ 0.05）.Tumor weight：The weight of tumor in control group was higher than other groups（P ＜0.01）,tumor inhibition rate in Exemestane group,Ruyanning group and combination group were 33.9％,25.2％,46.0％ respectively.The index of apoptosis in combination group was the highest,while in control group was the lowest.Comparing to control group,the difference was significant （P ＜ 0.01）.Conclusion：Ruyanning can enhance Exemestane effect to induce apoptosis,improve the rate of tumor inhibition on tumor-bearing nude mice.
