大鼠山仙颗粒含药血清对S-180肉瘤细胞bcl-2、VEGF表达的影响

Effect of rat serum containing Shanxian cranule on bcl - 2 and VEGF in S - 180 sarcoma cells

目的:通过观察山仙颗粒(SXG)含药血清对S-180肉瘤细胞bcl-2、VEGF蛋白表达的影响,探讨其治疗肿瘤性疾病的作用机制。方法:S-180肉瘤细胞分别与大鼠空白血清及含不同剂量SXG的药物血清在体外共同培养。于24 h、48 h、72 h后制备细胞涂片,分别用免疫荧光化学(FITC)法和免疫细胞化学(SABC)染色,检测不同浓度及不同作用时间的含药血清对S-180肉瘤细胞中bcl-2、VEGF蛋白表达的影响。结果:SXG含药血清可降低S-180肉瘤细胞中VEGF、bcl-2蛋白的表达,且呈浓度和时间依赖性。结论:大鼠SXG含药血清能抑制肿瘤细胞生长,其作用机制可能与其抑制S-180肉瘤细胞内VEGF的表达,下调bcl-2的表达有关。

Objective：To explore the mechanism of the serum containing SXG in treating neoplastic diseases by observing its effect on S-180 sarcoma cells bcl-2 and VEGF protein.Methods：S-180 sarcoma cells were co-cultured in vitro respectively with the rats blank serum and serum containing SXG of different doses;the cell smears were prepared in 24h,48h and 72h.The concentrations in different time were detected by immunocytochemistry （SABC） and immunofluorescence （FITC）.Results：Serum containing SXG can reduce the occurrence of bcl-2 and VEGF protein in S-180 sarcoma cells,and witha dose-time-dependent manner.Conclusion：Rats serum containing SXG can inhibit the growth of tumor cells,and its mechanism might be related to VEGF inhibiting and bcl-2 decreasing.
