摘要
目的:通过RNA干涉技术沉默p110β在前列腺癌细胞C4-2中的表达,观察p110β的表达抑制对前列腺癌增殖、侵袭、转移的影响。方法:根据p110β基因序列设计并合成的siRNA片段(p110β-siRNA)转染C4-2细胞,利用荧光实时定量qRT-PCR检测细胞中p110β基因mRNA水平的变化;Western blot检测p110β蛋白水平的变化;MTT法检测C4-2细胞增殖情况;Transwell小室侵袭实验检测C4-2细胞侵袭能力变化;细胞划痕试验检测对C4-2细胞迁移能力的影响。结果:p110β-siRNA下调了C4-2细胞中p110βmRNA水平与蛋白水平的表达,转染细胞的增殖和侵袭迁移能力下降。结论:p110β-siRNA能够下调p110β的表达,并有效抑制C4-2前列腺癌细胞的增殖及其迁移侵袭能力。因此,沉默p110β的表达可使前列腺细胞恶性程度降低。
Objective:To explore the effects of silencing p110β on proliferation and invasion and metastasis of huaman prostate cancer cell line C4-2.Methods:To design and synthesis of siRNA fragments (p110β-siRNA) according to p110β gene sequence and transfect p110β-siRNA into C4-2 cells;Using real time fluorescent quantitative qRT-PCR to detect the mRNA expression; Western blot technology to detect protein expression level ; the proliferation of C4-2 cells was analyzed by MTT ;Transwell cell invasion assay and wound scratch assay for detecting the invasion and metastasis ability of C4-2 cells.Results:p110β-siRNA down regulated mRNA and protein expression level of p110β gene in C4-2 cells and inhibited the proliferation,migration and invasion ability of C4-2 cells.Conclusion:p110β-siRNA could down regulate the expression of p110β,and effectively inhibit cell proliferation,migration and invasion abilities of C4-2 cells,p110β may be a target gene for the therapy of prostate cancer.
