Caveolin-1对血管吻合口白细胞介素10和白细胞介素6表达的影响 被引量:1

Effect of Caveolin-1 on the Expression of Interleukin-10 and Interleukin-6 in Vascu-lar Anastomotic Stoma

摘要 目的建立兔颈总动脉血管吻合口狭窄模型,并用局部转染小凹蛋白(Caveolin-1)质粒,观察血管吻合口部位Caveolin-1对白细胞介素10(IL-10)和白细胞介素6(IL-6)表达的影响。方法从南华大学动物中心取成年家兔32只,行颈总动脉血管吻合术,运用脂质体局部转染Caveolin-1质粒,随机分为4组:正常组(假手术组)、手术组、手术+空转染组、手术+转染组。分别于7天及28天取颈总动脉标本,通过HE染色观察内膜增生情况,并测量内膜与中膜面积比值(IA/MA);实时荧光定量PCR检测血管中Caveolin-1和IL-6、IL-10 mRNA的表达,免疫组织化学观察血管组织中IL-6、IL-10蛋白的表达。结果 HE染色显示,手术组及空转染组内膜明显增殖,管壁增厚,管腔缩小;而转染组未见明显内膜增殖及管壁增厚;从血管内膜/中膜面积比值发现,转染组比手术组明显降低;血管吻合手术后,抗炎因子IL-10和促炎因子IL-6都升高。当吻合口部位Caveolin-1基因高表达时,IL-10 mRNA和蛋白表达都上调,而IL-6 mRNA和蛋白表达都减少。结论 Caveolin-1促使血管吻合口部位炎症因子的平衡向抑炎因子倾斜。 Aim To investigate the effect of the caveolin-I on the expression of intedeukin-10 (IL-10) and inter- leukin-6 (IL-6) in rabbit carotid artery anastomotic stoma. Methods 32 New Zealand white rabbits were randomly di- vided into sham operation group, operation group, empty vector group and caveolin-1 transfected group. The rest speci- mens were taken for HE staining at 7 d and 28 d, which were used to observe the proliferation of intima, and the ratio of in- tima/media area was measured by Image-Pro Plus 6. 0 software ; Real time PCR was used to detect the mRNA, and immuno- histochemistry was used to assay the expression of protein. Results Anti-inflammatory eytokine IL-10 and pro-inflam- matory cytokine IL-6 were increased by carotid artery end-to-end-intestinal anastomosis. Local transfected caveolin-1 plas- mid can increase the expression of caveolin-I mRNA in rabbit carotid artery anastomotic stoma. Compared with other groups, the mRNA and protein of IL-10 were increased in caveolin-I transfected group. However, IL-6, a potent pro-in- flammatory cytokine, was decreased in caveolin-1 transfected group. Conclusion Caveolin-1 promotes anti-inflamma- tory factor in the balance between the pro- and anti-inflammatory cytokines in rabbit carotid artery anastomotic stoma.
