卡维地洛和心得安对门静脉高压症大鼠门静脉压力影响的研究

Carvedilol and propranolol on the portal vein pressure in rats with portal hypertension

目的观察比较卡维地洛和心得安对门静脉高压症大鼠门静脉压力的影响。方法采用CCl4加酒精制作大鼠门脉高压症模型,造模16周后用直接测压法测正常组、模型组大鼠门静脉压力,确定造模成功后分别给大鼠服用卡维地洛、心得安,并于2周、4周后用直接测压法测心得安组、卡维地洛组大鼠门静脉压力。结果模型组门静脉压力较正常组明显升高(P<0.01),卡维地洛组、心得安组门静脉压力较模型组明显下降(P<0.01),但仍高于正常组(P<0.01),2周卡维地洛组门静脉压力较心得安组无明显差异(P>0.05),4周卡维地洛组门静脉压力较心得安组下降(P<0.05)。结论卡维地洛可以改善门静脉系统的高动力状态,其治疗效果优于心得安,在肝硬化门静脉高压症的治疗中的作用值得进一步研究。

Objective To compare the effect of carvedilol and propranolol on the portal vein pressure in rats with portal hypertension. Methods The models of rats with portal hyperten- sion were prepared with CC14 and alcohol, the rats were separately given carvedilol and prop- ranolol after 16 weeks, and the portal vein pressure of the normal group, model group, prop- ranolol treatment group and carvedilol treatment group were measured by direct manometric method at 2 and 4 weeks. Results The portal vein pressure of model group was increased sig- nificantly compared with that of normal group （ P 〈 0. 01 ）, while such index in carvedilol treat- ment group and propranolol treatment group was higher than that of normal group （ P 〈 0. 01 ）, but was lower significantly than that of model group （P 〈 0.01 ）. There was no obvious differ- ence in portal vein pressure between carvedilol treatment group and propranolol treatment group after 2 weeks （ P 〉 0. 05 ）. After 4 weeks, the portal vein pressure of carvedilol treatment group was declined compared with that of propranolol treatment group （ P 〈 0.05 ）. Conclusion Carvedilol can alleviate the hyperdynamic state of portal system and is superior to the propranol- ol group, the effect in the treatment of the portal hypertension deserves further study.
