摘要
目的:观察元胡止痛胶囊对实验性偏头痛的治疗作用及可能的机制。方法:将大鼠分为空白组、模型组、中、西药阳性组和元胡止痛胶囊高、中、低剂量组,利用硝酸甘油制造偏头痛模型,分别以元胡止痛胶囊1.40、0.70、0.35g/kg灌胃5天,观察治疗后各组大鼠行为变化,酶联免疫法(ELISA)测定血浆中5-羟色胺(5-HT)、去甲肾上腺素(NE)、内皮素(ET)、降钙素基因相关肽(CGRP)及脑组织中5-HT、NE、多巴胺(DA)的含量。结果:模型组大鼠出现耳红、挠头、爬笼等现象,元胡止痛胶囊0.35、0.70、1.40g/kg各组现象不明显。硝酸甘油所致偏头痛模型大鼠血浆5-HT、NE、ET、CGRP含量较空白组均显著升高,脑组织中5-HT含量较空白组显著降低,NE、DA含量显著升高;与模型组比较,元胡止痛胶囊0.35、0.70、1.40g/kg均能使血浆5-HT、NE、ET、CGRP含量降低,脑组织中5-HT含量升高,NE、DA含量降低,且均有统计学意义。结论:元胡止痛胶囊能显著缓解模型大鼠因头痛所致耳红、挠头、爬笼等现象,具有调节大鼠5-HT、NE、DA、ET、CGRP等单胺类递质和血管活性物质含量的作用,这可能是其治疗偏头痛的作用机制之一。
Object: To observe the influence and action mechanism of Yuanhu Zhitong capsule on experimental migraine rats.Method: The rats were divided into control group,model group,three doses groups of Yuanhu Zhitong capsule(1.40,0.70,0.35g / kg),Chinese medicine and western medicine control groups.The rat model with migraine is build up by nitroglycerin.The change of behavior was detected and the level of neurotransmitter such as 5-HT in brain and blood plasma of rat is tested with ELISA.Result: Red ears appeared,the frequency of scratching head and climbing cage frequency increased in model group,the treatment group of 0.35,0.70,1.40g / kg is not obvious.The content of 5-HT,NE,ET,CGRP in blood plasm of model rat is increased.The content of 5-HT in brain is decreased and NE,DA in brain is increased.Yuanhu Zhitong capsule of 0.35,0.70,1.40g / kg can decrease the content of 5-HT,NE,ET,CGRP in blood plasma of model mice,can also increase the content of 5-HT in brain and decrease the content of NE,DA in brain of model mice.Conclusion: Yuanhu Zhitong capsule would have effects on migraine model.
