摘要
目的:研究复方参丹芎注射液对大鼠局灶性脑缺血再灌注损伤的保护作用。方法:采用线栓法堵塞大鼠大脑中动脉(MCAO)脑缺血/再灌注损伤模型(MCAI/R),将造模成功的雄性大鼠随机分为假手术组、模型组、参丹芎注射液高、中、低剂量组(20、10、5mg提取物/kg)、金纳多组(35mg提取物/kg),分组后立即iv给药,连续给药3d后考察神经功能缺失症状,脑梗死灶面积,脑组织中丙二醛(MDA)含量、乳酸脱氢酶(LDH)、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活性的变化。结果:参丹芎注射液(20、10mg提取物/kg)能够明显减轻脑缺血/再灌注损伤大鼠神经功能缺损症状,减少脑梗死灶面积和MDA含量、增加LDH、SOD和GSH-Px活性。结论:参丹芎注射液对大鼠脑缺血再灌注损伤具有明显的保护作用。
Objective: To study the protection of compound Shendanxiong injection on rat brain after cerebral ischemia-reperfusion injury.Methods: Rat model of cerebral ischemia-reperfusion(MCAI /R) injury was created by the middle cerebral artery occlusion(MCAO) with suture method.MCAI / R-established male SD rats were randomized into sham control group,model group,high-,middle-,and low-dose(20,10 and 5mg / kg) of shendanxiong injection groups,and positive control group(treated with extract of Ginkgo Biloba leaves injection 35mg / kg).Changes in symptoms of neurological deficit,cerebral infarction area,content of MDA and activities of LDH,SOD,GSH-Px in brain were measured after rats were iv administrated with shendanxiong injection for three days.Results: Shendanxiong injection(20,10mg / kg) could significantly improve symptoms of neurological deficit,reduce cerebral infarction area and MDA content,increase the activities of LDH,SOD and GSH-Px.Conclusion: Shendanxiong injection has significant protective effect on cerebral ischemia-reperfusion injury in rats.
