雷公藤多苷对变应性接触性皮炎患者趋化因子IP-10、Eotaxin mRNA表达的影响 被引量:2

摘要 目的观察雷公藤多苷含药血清对变应性接触性皮炎患者(allergic contact dermatitis,ACD)趋化因子γ干扰素诱导蛋白10(interferon-gamma-inducible protein-10,IP-10)、嗜酸性粒细胞趋化因子(Eosinophil chemoattractant chemokine,Eotaxin)mRNA表达的影响,进一步探讨雷公藤多苷治疗接触性皮炎的机制。方法逆转录聚合酶链式反应(Reverse Transcription Polymerase,RT-PCR)方法测定与不同浓度雷公藤多苷共同培养ACD患者外周血单一核细胞(PBMC)中IP-10、Eotaxin mRNA的表达。结果雷公藤多苷高、中、低3个浓度组体外培养的ACD患者PBMC中IP-10 mRNA表达明显低于对照组,差异均有统计学意义(P<0.01);雷公藤多苷含药血清3个浓度组之间比较,IP-10mRNA表达差异无统计学意义(P>0.05);雷公藤多苷含药血清3个浓度组之间及与对照组比较,体外培养的ACD患者PBMC中Eotaxin mRNA的表达差异均无统计学意义(P>0.05)。结论雷公藤多苷治疗ACD的机制之一可能是通过调节ACD患者PBMC中趋化因子IP-10、Eotaxin mRNA的水平,改变细胞因子的表达,促进Th1/Th2平衡来实现的。 Objective To study the effect of Tripterygium Wilfordii Polyglycosidium (TWP) on allergic contact dermatitis (ACD) by detect the expression of chemokine interferon-gamma-inducible protein-10 (IP-10) and Eosinophil chemoattractant chemokine (Eotaxin) messenger ribonucleic acid(mRNA). Methods Reverse Transcription Polymerase(RT-PCR) was used to detect the mRNA of IP-10 and Eotaxin in PBMC of ACD curtured with different concentration TWP. Results Compared with the control group, three different concentration groups (5.0, 1.0, 0.5 mg/L) of TWP all could remarkably suppress the expression of IP-10 mRNA in PBMC of ACD (P〈0.01). While there was no significant difference among these three different concentration groups of TWP in there suppression on the expression of Eotaxin mRNA in PBMC of ACD (P〉0.05). There was no significant different among the three different concentration groups of TWP on the expression of Eotaxin mRNA in PBMC of ACD (P〉0.05). Conclusion TWP could regulate the expression of IP-10 and Eotaxin mRNA as well as eytokines expression ratio so as to modulate the Thl/Th2 balance, which may be one of the mechanisms of TWP treatment on ACD.
