调气活血补肾汤对慢性肾衰竭大鼠肾功能的影响 被引量:1

摘要 目的:探讨调气活血补肾汤对慢性肾功能衰竭(CRF)大鼠肾功能、肾组织一氧化氮(NO)及脂质过氧化的影响。方法:Wistar大鼠64只随机分为模型组、调气活血补肾汤大剂量治疗组、小剂量治疗组和正常对照组,前3组大鼠麻醉后,用摘除5/6肾脏皮质的方法制备CRF模型;模型制备后,小剂量模型组给予调气活血补肾汤生药13.9 g/kg灌胃,大剂量组给予27.8 g/kg灌胃,模型组给予蒸馏水,各组给药4周、10周分别处死8只大鼠,测定肾功能及NO含量、一氧化氮合酶(NOS)及超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量。结果:大鼠CRF模型成功制备,调气活血补肾汤大剂量组大鼠血清尿素氮(BUN)、肌酐(Cr)明显降低;CRF模型组4周时匀浆NO水平和NOS活性均较对照组明显升高,随时间延长呈下降趋势,大剂量组、小剂量组用药4周NO水平和NOS活性下降,10周时持续降低;且治疗后肾组织SOD活性升高,MDA水平下降,大剂量组均优于小剂量组。结论:调气活血补肾汤可改善肾脏功能,其机制与其调节肾功能NO的代谢平衡、降低自由基损伤有关。 Objective: To investigate the influence of Tiaoqihuoxuebushen decoction on kidney function, kidney NO and lipid peroxidation of Wistar rats with chronic renal failure (CRF). Methods: Sixty-four Wistar rats were randomly divided into model group (group M), large-do se treatment group (group L), small-dose treatment group (group S) and normal control group (group C) with 16 in each. After anesthesia, CRF model were made in the first three groups. Rats in group S were given 13.9 g/ kg of Tiaoqihuoxuebushen decoction via intragastric administration, while those in group L were given 27.8 g / kg of the decoction. Distilled water was given to group M. In 4 w and 10 w after drug administration, 8 rats were executed respectively, and renal function, NO, NOS, SOD and MDA concentrations were measured. Results: CRF model was established successfully, and in group M, NO and NOS activities elevated remarkably than group C in early stage, and decreased along with time. Serum BUN and Cr concentrations in cRF rats were lowered significantly. In groups L and S, NO and NOS activities decreased in 4 w, and kept decreasing in 10 w after treatment, but were still higher than those in groups M and C. Differences between groups were statistically significant (P 〈 0.05 ). Besides, SOD activities were enhanced significantly after treatment, and decreased in 10 w later. The influences mentioned above were better in group L than those in group S. Conclusion: Tiaoqihuoxuebushen decoction can improve kidney function. Its mechanism is related to lowering free radical injury by enhancing NOS activity and promoting NO composition.
