摘要
对从长白山温泉泥样中分离出的高温菌的培养条件进行研究,考察培养温度、培养基起始pH、装液量、摇床转速对其生长情况的影响。在单因素试验的基础上,通过正交试验优化,确定该菌株的最佳培养条件为:培养温度为65℃、起始pH 6.5、装液量45mL/100mL、摇床转速为150r/min,在此条件下菌株的生物量OD600是1.369,活菌数为2.1×108cfu/mL。
We studied the culture conditions of the thermophilic bacteria isolated from the mud samples in Changbai Mountain Springs,and researched the influence of cultivating temperature,initial pH of medium,incubation time,and rotating speed on the growth of strains.Based on the single-factor tests,the culture conditions of strains were optimized by orthogonal experiment.Results showed that the best cultivate temperature was 65℃,the initial pH was 6.5,loaded liquid was 45mL /100mL,speed of rotary shaker was 150 r / min,and the biomass OD600 of strains was 1.369 and the number of viable cells was 2.1 × 108cfu / mL under these conditions.
