摘要
为了获得生物利用率高且生物安全性良好的补锌制剂,本文采用水体系合成法制备类人胶原蛋白(human-like collagen,HLC)-锌螯合物,考察pH值和配位比对锌螯合率的影响,并通过紫外—可见吸收光谱和傅立叶红外光谱研究该锌螯合物。优化的最佳工艺条件为:体系pH值6.0且类人胶原蛋白与ZnSO4的质量比10∶4。光谱研究结果表明,类人胶原蛋白和Zn2+之间存在相互作用,生成的锌螯合物是一种不同于HLC的新化合物;羰基和亚氨基均参与HLC-Zn螯合物的形成过程,且该螯合物保持HLC分子原有的α-螺旋结构。因此,本文的研究结果为类人胶原蛋白—锌螯合物的大规模制备和进一步研究开发做出了有益探索。
The complex of human-like collagen(HLC) with zinc was prepared in aqueous solution and was analyzed by UV-vis spectroscopy and FTIR.The effects of pH value and reactant ratio on the coordination efficiency were studied,and the optimum conditions were the pH value of 6.0 and the mass ratio of HLC to zinc sulfate 10∶4.In addition,it could be concluded from UV-vis spectra that there existed interaction between HLC and zinc,and the complex was a new chemical compound different from pure protein.In the complex of HCl-Zn,human-like collagen acted as ligand and was linked with the zinc ions via both groups of C = O and N-H,and the resultant complex retained the helical structure of HLC.The results provided a useful guideline for the large-scale preparation and further research of the HLC-Zn complex.
