摘要
目的:探讨 HSP70单抗及反义寡核苷酸对前列腺癌细胞株 LNCaP和 PC 3m凋亡的诱导作用及差异。方法:用流式细胞术分析了 HSP70单抗( 2μ l)、反义寡核苷酸( 10μ mol/L)及单抗联合反义寡核苷酸分别处理 LNCaP和 PC 3m 24、 48、 72、 96 h后,细胞凋亡的发生率。结果:在 48、 72、 96 h时单抗联合反义寡核苷酸诱导的凋亡发生率高于单抗或反义寡核苷酸单独诱导时的凋亡率,差异显著 (分别为 P0.05);反义寡核苷酸与单抗在 48、 72、 96 h诱导的凋亡率间差异显著 (P0.05)。结论: HSP70单抗与 HSP70反义寡核苷酸联合可以显著诱导前列腺癌细胞 LNCaP和 PC 3m发生凋亡,为生物疗法治疗前列腺癌提出了新途径。
Objective: To study apoptosis induced by HSP70 monoclonal antibody(MAb) and antisense oligomer and its difference between prostatic cancer cell line LNCaP and PC 3m. Methods: Apoptosis of LNCaP and PC 3m,which were treated by HSP70 MAb(2μ l), antisense oligomer (10μ mol/L) and HSP70 MAb combining with antisense oligomer for 24, 48, 72, 96 h respectively,were analysed by flow cytometry. Results: Apoptotic rates of LNCaP and PC 3m induced by HSP70 MAb combining with antisense oligomer in 48, 72, 96 h were higher than those by HSP70 MAb or HSP70 antisense oligomer, With significant differences(P0.05).
