摘要
采用SSR分子标记的方法,对70份玉簪种质资源进行了遗传多样性分析,筛选出合适的提取DNA的方法,以及扩增效果较好的5对引物。结果表明:根据SSR结果得到的遗传图谱显示70种玉簪的相似系数在0.66~O.93之间。在遗传系数为0.73处,可以将它们分为三大类:第一大类包括1个品种H.Krossaregal,它是优良的蓝灰叶品种;第二大类包括H.LoverPat、H.Pizzaz、H.Captainkirk、H.FrancesWilliama、H.E1egans、H.9F:86、H.Kroea、H.Ventricosa、H.So.sweet、Hmentreal、H.RegalSpender、H.Nightfortellnei、H.SunPower、H.August.moon、H.Dianaremember,它们都属于大中型玉簪;其余的40个品种为第三大类,它们大都生长快速,是国内外比较受欢迎的品种。由于试验材料都采摘于同一个地区,生长的地理环境相同,所以它们之间的相似系数比较高但由于品种不一样,所以还是存在着一定的差异性。
Using SSR molecular marker method, the genetic diversity of 70 Hostas were analyzed and the suitable method of DNA extraction and the best effect amplifid primers were selected. The results showed that from the picture of Genetic relationship of 70 materials from SSR data that the whole Hosta similarity coefficient between 0. 66~0. 93. The genetic coefficients for 0. 73 place,they could be divided into three categories. The first class includes H. Krossa regal,it was the best variety of Blue ash leaf;the second class includes H. Lover Pat, H. Pizzaz, H. Captain kirk, H. Frances Williama, H. Elegans,H. 9F:86,H. Kroea, H. Ventricosa, H. So. sweet, H. mentreal, H. Regal Spender, H. Night for tellnei, H. Sun Power,H. August. moon, H. Diana remember,they all belonged to the large and medium-sized Hostas;the remaining 40 varieties belonged to the third part, most of them grow fast, they were also being the domestic and foreign popular varieties. Because the experimental materials were harvested in the same area, the growth of the geographical environment was identical, so the similarity coefficient between them was relatively high but because of different varieties so there were still some differences.
