摘要
目的建立葫芦素胶束药物含量测定。方法采用Hypersil ODS C18柱(4.6×200 mm,5μm),流动相为乙腈-水-磷酸(45∶55∶0.1,v/v),检测波长228 nm,流速:1.0 ml/min。结果葫芦素在0.25~20.0μg/ml的浓度范围内,峰面积对浓度有良好的线性关系,回归方程为:A=26823×C+866.89(R2=0.9999,n=7),回收率分别为99.11%、99.52%、100.5%,方法的日内与日间精密度RSD均<2%。结论该方法具有简便、快速、准确的特点,可用于葫芦素胶束药物含量测定。
Objective To develop the method of high performance liquid chromatograghy(HPLC) which will determine the contents of Cucurbitacin.Methods The separation was performed on a Hypersil ODS C18 column(24.6 × 200mm,5μm) at room column temperature.The mobile phase consisted of acetonitrile-waterphosphoricacid(45∶ 55∶ 0.5) at the flow rate of 1ml.min^-1.The detection wavelength was at 228 nm.Results The linear range for cucurbitacin was in the range of 0.25 ~ 20.0μg.ml-1(R2 = 0.9999,n = 7).The withinday and between-days variation coefficients were less than 2%.The average recoveries were 99.11%,99.52% and 100.5% respectively.Conclusions A simple,rapid and accurate method for determination cucurbitacin in micelle is established.
