摘要
对分离自威海近海海域的一株产红色素的海洋放线菌SHXF02-1进行了红色素提取方法及其稳定性研究,测定其最大吸收波长,考察了pH值、温度、光照、氧化剂和还原剂、防腐剂等因素对色素稳定性的影响。此外,对菌株SHXF02-1进行了分子鉴定,利用PCR技术扩增了其16S rDNA序列,利用软件MEGA4.1构建了系统进化树。实验结果表明,该红色素在波长530nm处有最大吸收峰,用乙醇浸提该色素优于其他试剂。该红色素为脂溶性色素,酸性条件下稳定,该色素的光、热稳定性好,耐氧化能力强,耐还原性较差。防腐剂食盐和蔗糖对色素均无不良影响。16S rDNA序列分析和系统进化树显示菌株SHXF02-1与多株micrococcus sp.的16S rDNA具有99%的相似性,初步鉴定菌株SHXF02-1是微球菌属Micrococcus sp.的一种。菌株SHXF02-1具有较高的研究和开发价值。
Extraction conditions, basic properties and stability of the red pigment produced by a marine actinomycete strain SHXF02-1, which was isolated from Weihai coastal sea area, were investigated in this paper. The maximum absorption wavelength was measured. It was tested that the effects of pH value, temperature, light, UV, oxidant, reducer, and preservatives on stability of the pigment. In addition, 16S rDNA of the strain SHXF02-1 was amplified by PCR for molecular characterization, and the software MEGA 4.1 was used to construct phylogenetic tree. The results showed that the red pigment had the maximum absorption value at 530 nm. Dissolubility of this pigment in the seven organic solvent hadbeen accessed, and the ethanol was optimally chosen for extraction. This red pigment was stable under the acidic condition, but sensitive to reducing agent, while had a good resistance to UV, light, heat and oxidant. Some common preservatives, such as salt and sucrose, had no adverse effects on stability of the pigment. Phylogenetic tree analysis of 16S rDNA sequence of SHXF02-1 indicated that the similarity with multiple strains of micrococcus sp. was all above 99%. Therefore, SHXF02-1 was identified as a new strain belonging to Micrococcus sp.
