Gel permeation chromatography clean-up for the determination of benzo (a) pyrene in edible vegetable oils by high performance liquid chromatography with fluorescence detector

摘要 建立一种凝胶渗透色谱-高效液相色谱法测定食用植物油中苯并(a)芘含量的方法。样品经凝胶渗透色谱净化处理,采用反相C18色谱柱Phenomenex Luna C18(4.6mm×250mm,5μm),以乙腈:水=91:9溶液为流动相,流速1mL/min,荧光检测器,激发波长384nm,发射波长406nm,对食用植物油中的苯并(a)芘进行检测。在2.0、10.0、25.0μg/kg添加量范围内,平均回收率为98.7%,相对标准偏差为3.4%(n=6)。方法的检出限为0.3μg/kg,线性范围为0.1~20.0μg/L(R=0.9992),测定结果与标准方法基本相同。本法快速、简便、准确,可以作为食用植物油中苯并(a)芘的检测方法。 This study developed a reverse phase HPLC method for determing benzo (a) pyrene in edible vegetable oils. After samples were pretreated by gel permeation chromatography, benzo (a) pyrene was separated on a Phenomenex Luna C18 (4.6 mm×250 mm, 5 μm) using a mobile phase composed of acetonitrile and H20 (91:1, v/v) at a flow rate of 1 mL/min and detected using a fluorescence detector set at λex=384 nm, λem=406 nm. At the spike levels of 2.0, 10.0, 25.0 μg/kg, the average recovery for benzo (a) pyrene was 98.7% with the RSD of 3.4% (n=6), the limit of detection was 0.3 pg/kg, and the linear range was from 0.1-20.0 μg/L (R=0.9992). The results of benzo (a) pyrene determination in four different edible vegetable oils samples by this method were almost the same as those determined by GB/ T 22509--2008. The method was fast, easy and accurate. Therefore, the method developed can be use to determine benzo (a) pyrene in edible vegetable oils.
