摘要
采用PCR和RACE的方法,获得大蒜果聚糖水解酶(FEH)基因的全长cDNA序列,并对其基因结构和系统进化进行分析。结果表明,所获得的cDNA序列全长1900bp,开放阅读框(ORF)为1668bp,编码555个氨基酸。在该氨基酸序列中发现已知FEH氨基酸序列的保守功能域;与小麦(Triticum aestivum)、黑麦(Lolium perenne)的FEH氨基酸序列的同源性为53%。基本确定首次获得了大蒜FEH基因。
To obtain a full cDNA sequence of a fructan exohydorolase (FEH) from galic (Allium sativum L.), PCR and RACE method was used.Genetic structure and phylogenetic analysis was done.The results showed that sequence length was 1900bp,in which the encoding region was 1668bp.The sequence encoded a polypeptide of 555 amino acids,in which deduced amino sequence of known FEHs was found.The amino sequence analysis showed 53% similarity to the FEHs amino sequence of Triticum aestivum and Lofium perenne.
