摘要 目的 :研究胶质细胞源性的神经营养因子 (GDNF)对面神经损伤后的促再生作用。方法 :家兔面神经双侧造成 5 mm神经缺损 ,以硅胶管套接后将硅胶管与神经外膜缝合固定。一侧硅胶管内注入纯化 GDNF,另一侧注入等量生理盐水对照。于不同时间段取材 ,切片行甲苯胺兰、HE、Bielschowsky银染改良法染色观察面神经元细胞体及再生神经结构。图像分析测量再生神经轴突直径及单位面积轴突数量、再生轴突恢复率。6 0 d及 90 d作透射电镜观察轴突及髓鞘超微结构并测定神经传导速度 (NCV)。结果 :与对照组相比 ,GDNF可以 :1保护面神经元细胞 ,减少面神经元的死亡数目 ;2提高轴突再生的数量及质量 ,并可早期恢复神经传导速度。结论 :面神经损伤后应用外源性 ?Objective: To study the effect of glial cell line derived neurotrophic factor (GDNF) on facial nerve regeneration after injury.Methods:Defect of 5 mm was made in each facial nerve on both sides in 32 rabbits.Silicon tubes were used to bridge the defects.Pure GDNF (15 μg in 30 μl silane) was injected into silicon tube on one side and saline (30 μl) on the other side as the control.Nerve regeneration was studied by morphological observation with HE and Bielschowsky stainning,counting of nerve neurons and axon and measuring of motor nerve conduction velocity.The rest 3 rabbits was as blank contol. Results:7 d and 17 d after operation ,the neuron number in GDNF treated and control groups were 39.2±2.3 and 28.4±1.9,and 33.6±2.1 and 27.1±1.4,respectively ( P <0.01).90 d after operation the axon number in the two groups were 2 836±621 and 1 564±386,respectively ( P <0.01),the motor nerve conduction velocities were 28.4±3.2 and 21.5±2.8, respectively ( P <0.01). Conclusion: GDNF can enhance the regeneration of facial nerve injured.
