摘要
目的 探讨氧化应激对肺泡表面活性物质结合蛋白A(SPA)表达的调控作用。 方法 提取SPA转录因子 (TTF1 ) ,采用凝胶电泳迟滞试验 (EMSA)及核转录试验 ,测定氧化应激对TTF1 与SPA基因调控区DNA的结合活性及SPA核转录活性。 结果 过氧化氢 (H2 O2 )剂量依赖地抑制TTF1 与SPA基因调控区DNA的结合及抑制SPA转录活性。 结论 氧化应激对TTF1 活性的抑制可能是急性肺损伤SPA表达下降的重要原因。
Objective To investigate the transcriptional regulation of oxidative stress on the expression of surfactant protein A (SPA). Methods The effect of H 2O 2 on DNA binding activity and SPA nuclear transcription activation in the SPA transcription factor TTF 1 and in regulating area of SPA gene were studied with electrophoretic mobility slow test (EMSA) and nuclear transcription test in vitro. Results H 2O 2 could inhibit the DNA binding activity and the SPA nuclear transcription activation dose-dependently in the TTF 1 and SPA gene regulating area. Conclusions Oxidative stress might be the important factor of the SPA down-expression regulation after acute lung injury.
