摘要
目的 研究顺铂对抑癌基因wtp53及癌基因C -myc在肝癌细胞凋亡过程中表达的影响。方法 用抑癌药顺铂诱导凋亡 ,通过细胞形态学检查 ,DNA琼脂糖凝胶电泳证明凋亡发生 ,采用原位杂交技术检测wtp53、C myc基因在人肝癌细胞株SMMC 772 1凋亡过程中的表达。结果 wtp53基因在顺铂作用后 1h开始表达 ,6h达到高峰 ,12h则明显下降。C myc基因在顺铂作用后表达逐渐增强 ,12h达到高峰。结论 wtp53、C myc基因均参与了肝癌细胞的凋亡过程 ,具有促进细胞凋亡作用。wtp53基因对C myc基因在转录水平上可能有上调作用。
Objective To study the expression of antioncogene wtp53 and oncogene C_myc in the apoptotic process of hepatocellular carcinoma cells.[WT5”HZ]Methods [WT5”BZ]The expression of wtp53 and C_myc gene in the apoptotic process were detected by using in situ hybridization after the cisplatin_induced apoptosis had been ascertained by morphology under electron and light microscopy,DNA gel electrophoresis. [WT5”HZ]Results [WT5”BZ]wtp53 gene mRNA expressed after exposing SMMC_7721 cells to cisplatin for 1 h and then the peak appeared in 6 h,but after 12 h the wtp53 mRNA level would decreased significantly.C_myc gene mRNA began to expressed after exposing SMMC_7721 cells to cisplatin for 1 h and then strongened gradually,and then the peak appeared in 12 h.[WT5”HZ]Couclusion [WT5”BZ]wtp53 gene may induced cell apoptosis,it played an important role in the process of hepatocellular carcinoma cells.wtp53 gene may up_regulate the expression of C_myc gene in the process of apoptosis.
