摘要
为探究热激对拟南芥热激因子AtHsfA1a表达的影响,对过量表达拟南芥热激因子AtHsfA1a的植株进行39℃热激5 min后提取其RNA,应用RT-PCR技术对热激因子AtHsfA1a的RNA进行逆转录和扩增,并对其进行电泳检测.结果表明:过量表达拟南芥热激因子AtHsfA1a的AtHsfA1a在热激和常温下表达量均比野生型高;而且野生型拟南芥和过量表达拟南芥热激因子AtHsfA1a的AtHsfA1a在39℃热激下的表达量比25℃常温下的高,说明AtHsfA1a的表达受热激诱导,结果可为研究拟南芥热激因子AtHsfA1a作用机理提供科学依据.
In order to research the effect of heat shock on the expression of the Arabidopsis heat shock factor AtHsfA1 a, Transgenic Arabidopsis of over expression AtHsfAla was used as experimental material. At first, heat shock factor AtHsfAla RNA was extracted un- der heat shock for 5 min, then reverse transcription and amplification of heat shock factor AtHsfA1 a RNA were preformed by RT-PCR and analyzed by agarose gel electrophoresis. The results showed that the AtHsfAla expression of transgenic Arabidopsis was higher than wild type under heat shock and at room temperature. AtHsfA1 a expression of transgenie Arabidopsis and wild type at 39 ~C were higher than 25 ~C. These indicated the expression of the Arabidopsis AtHsfA1 a was induced by heat shock. This study will lay the foundation to study the mechanism of the Arahidopsis heat shock factor AtHsfA1 a.
