原子力声显微镜评价大鼠心肌骨架

摘要 目的本研究用原子力声显微镜观察大鼠心肌细胞的细胞骨架,并对声像图进行分析。方法使用原子力显微镜加声激励对在盖玻片上贴壁生长的H9C2大鼠心肌细胞细胞骨架成像。结果使用原子力声显微镜可以完整显示H9C2大鼠心肌细胞的细胞膜、细胞核以及清晰显示网状结构的细胞骨架结构(微丝、微管、中间丝)。结论原子力声显微镜可以很好地显示大鼠心肌细胞表面及亚表面的细胞骨架结构,进而为研究病理状态的心肌细胞打下基础。 Objective To visualize the rat cardiomyocytes and cell structure with atomic force acoustic microscopy(AFAM).Methods Rat cardiomyocytes H9C2 were cultured on cover slips.H9C2 cells on cover slips were subject to acoustic excitation,and scanned with AFAM Veeco DI 3100,the images were processed with Thermomicroscopes Proscan Image Processing software version 2.1.Results Cellular structure including cell membrane,three-dimensional cytoskeleton network,morphology of cell nucleus and intercellular conjunction of rat cardiomyocytes were clearly and accurately displayed using AFAM.Conclusion Cell structure and morphology of rat cardiomyocytes can be visualized using atomic force acoustic microscopy.
