摘要
目的 观察 1,2 5二羟维生素D3 [1,2 5 (OH) 2 VD3 ]对培养兔骨髓细胞产生立即早期基因c -fos的蛋白产物 (FOS蛋白 )的诱导作用。方法 在原代培养至第六代的兔骨髓细胞中加入含 5× 10 -5M/L 1,2 5 (OH) 2 VD3的培养液 ,继续培养 3小时后 ,固定标本 ,进行FOS免疫组化反应。结果 大部分细胞形态正常 ,胞核呈棕黄色 ,椭圆形 ,胞浆不着色 ,细胞轮廓清晰 ,表现为FOS免疫组织化学反应阳性。结论 1,2 5 (OH) 2 VD3 能够诱导兔骨髓细胞表达FOS蛋白 。
Objective To observe the induction of FOS protein by 1,25(OH) 2VD 3 in cultured rabbit bone marrow cells.Methods Adding culture fluid containing 5×10 -5 M/L 1,25(OH) 2VD 3 into the cultured sixth passage rabbit bone marrow cells,after co-culturing for 3 hours,FOS immunocytochemical reaction was conducted.Results Most of the nuclei of bone marrow cells were stained dark yellow-brown with no stain in cytoplasm and were FOS positive immunocytochemical reaction.Conclusion It is suggested that 1,25(OH) 2VD 3 might play an important role in the process of bone remodeling through c-fos signal transduction passway.
