HIV慢性感染者Vδ2T淋巴细胞的CD25表达与其凋亡及疾病进展的关系研究

Correlations between CD25 expression on Vδ2 T lymphocytes and the apoptosis of Vδ2 T lymphocytes and disease progression in chronically HIV-infected patients

摘要 目的探讨HIV慢性感染者Vδ2T淋巴细胞上CD25的表达与其频率、凋亡及HIV感染疾病进展的关系。方法选择本院2012年3月—2013年4月门诊随访的HIV慢性感染者31例及健康对照者10例。31例HIV慢性感染者中,未接受高效抗反转录病毒治疗(highly active antiretroviral therapy,HAART)的患者10例(未治疗组),治疗3个月的患者10例(治疗3个月组),治疗6个月的患者11例(治疗6个月组)。应用流式细胞术检测Vδ2 T淋巴细胞频率、凋亡率及CD25表达情况,观察未治疗组、治疗3个月组和治疗6个月组上述指标的变化,分析这些变化与CD4+T淋巴细胞计数和病毒载量的关系,并分析CD25表达与Vδ2 T淋巴细胞频率和凋亡率的相关性。结果 HIV慢性感染者Vδ2 T淋巴细胞上CD25表达与健康对照者相比显著增加,给予HAART 6个月可使其下调,但仍高于健康对照者。HIV慢性感染者Vδ2 T淋巴细胞上CD25表达具有随CD4+T淋巴细胞计数增加而下降的趋势,但相关性无统计学意义;其与未接受HAART患者的病毒载量相关性不显著。HIV慢性感染者Vδ2 T淋巴细胞频率与其CD25表达呈负相关,但其凋亡率与CD25表达相关性不显著。结论 HIV慢性感染者Vδ2 T淋巴细胞处于免疫激活状态,其异常免疫激活可能与其频率下降有关。6个月的HAART可部分降低其激活水平和凋亡率,但不能使其频率恢复。 Objective To investigate the correlations between CD25 expression on Vδ2T lymphocytes and the frequency and apoptosis of Vδ2T lymphoeytes and disease progression in chronically HIV-infected patients. Methods Thirty-one chronically HIV-infected outpatients treated in our hospital from Mar. 2012 to Apr. 2013 and 10 healthy controls (HC) were enrolled in the study. Of 31 chronically HIV-infected patients, 10 didn't receive highly active antiretroviral therapy (HAART) (without HAART group), 10 received 3 months of HAART (3-month HAART group), and 11 received 6 months of HAART (6-month HAART group). The frequency and apoptosis of Vδ2T lymphocytes and CD25 expression on Vδ2T lymphoeytes were detected by flow cytometry. The changes of the above-mentioned indicators of without HAART group, 3-month HAART group and 6-month HAART group were observed respectively, and the correlations between these indicators and CD4^+ T lymphocyte counts and viral loads of each group were analyzed respectively. The correlations between CD25 expression and the frequency and apoptosis of Vδ2T lymphocytes were analyzed as well. Results CD25 expression on Vδ2T lymphoeytes in chronically HIV-infected patients increased significantly as compared with HC. Though 6 months of HAART could down-regulate it, the expression was still higher than that of HC. CD25 ex- pression on V82 T lymphocytes decreased along with the increase in CD4~ T lymphocyte counts, but the correlation was not signifi- cant. The correlation between CD25 expression on V82 T lymphocytes and viral loads in the patients not receiving HAART was not significant. The frequency of V82 T lymphocytes was negatively correlated with CD25 expression in chronically HIV-infected pa- tients, but the correlation between the apoptosis of and CD25 expression on Vδ2T lymphocytes was not significant. Conclusions Vδ2T lymphoeytes are activated in chronically HIV-infected patients, which may be correlated with the decline in the frequency of Vδ2T lymphocytes. Six months of HAART can reduce the activation level and the apoptosis partly, but can't restore the frequency of Vδ2T lymphocytes.
