中国林蛙凝集素样核糖核酸酶A的抑菌作用

摘要 目的:从中国林蛙基因组中克隆核糖核酸酶的基因并分析该基因的特殊生物学功能。方法:采用PCR方法克隆基因,利用原核系统进行基因表达,检测重组蛋白的抑制细菌生长的活性。结果:DNA顺序分析显示:克隆的基因编码了111个氨基酸序列,其中拥有核糖核酸酶A家族的保守序列,与日本稻田蛙的一种凝集素样核糖核酸酶的氨基酸相似性达到82%,与中国林蛙Onconase样核糖核酸酶氨基酸序列仅有40%相似性。重组蛋白对革兰氏阴性菌和革兰氏阳性菌均有生长抑制作用。结论:中国林蛙凝集素样核糖核酸酶有抑制细菌生长的作用。 Objective:A ribonuclease A gene will be cloned from Chinese frog (Rana Dybowskii) genome and the special function will be investigated. Method:PCR method was used for gene cloning,prokaryotic system for gene expression and the function of the recombinant protein was analyzed for antibacterials. Result:The DNA sequence analysis for cloned gene showed that the gene cedes 111 amino acid se- quence for the mature protein and possesses the conserved sequences in ribonuclease A superfamily. The identity to lectin from Japanese frog (Rana japonica) is up to 82%, and only 40% to Onconase - like ribonuclease found in same species. The recombinant protein showed inhibitory effect on the bacteria growth. Conclusion: The lectin - like ribonuclease from Rana Dybowskii has a function to inhibit the growth of the bacteria.
