Nedd化参与调节泛素化进程,与多种凋亡相关因子及细胞周期蛋白的降解密切相关。Nedd化及去Nedd化受多种催化酶的精密调控,这些酶蛋白的异常表达会导致细胞内Nedd化增强或减弱,从而影响细胞内蛋白的平衡,与黑素瘤的发生密切相关,Nedd化在肿瘤的发生发展中起重要作用,该通路的多个环节有希望成为新的抗肿瘤药物的作用靶点。
Neddylation is involved in protein ubiquitination, and closely associated with the degradation of many cell cycle proteins and apoptosis-related factors. Neddylation and deneddylation are delicately regulated by a series of enzymes. Abnormal expression of these enzymes would cause attenuation or enhancement of neddylation, which may break the balance between intracellular proteins, and finally induce the development of melanoma. Many molecules in the neddylation pathway are expected to be novel targets of antitumor agents.
