摘要
目的探讨TGF-β1在大肠侧向发育型肿瘤中的表达情况及其临床意义。方法对于12例经内镜下黏膜切除的侧向发育型肿瘤(laterally spreading tumor,LST)组织、12例腺瘤性息肉组织标本以及16例经外科手术切除的大肠癌组织标本采用实时荧光定量PCR(qRT-PCR)法检测TGF-β1基因表达,蛋白印迹(Western blotting)法检测TGF-β1蛋白表达,并分析其在大肠不同病变中差异性表达的意义。结果 qRT-PCR结果提示,TGF-β1 mRNA在侧向发育型肿瘤组织中表达水平显著高于腺瘤性息肉组织,但明显低于大肠癌组织。Western blotting结果显示TGF-β1在正常肠黏膜及腺瘤性息肉组织中几乎不表达,而在大肠癌中表达量明显升高。LST组织中的TGF-β1蛋白表达量较正常肠黏膜及腺瘤性息肉高,但明显低于大肠癌组织。结论 TGF-β1在LST中的表达水平明显高于正常肠黏膜及腺瘤性息肉组织,同时又与大肠癌组织中的显著高表达有所区别,提示其可能成为判断LST恶变倾向及指导随访治疗方案的重要参考指标。
Objective To explore the mRNA and protein expression of TGF-β1 in colorectal laterally spreading tumor (LST) , and investigate its clinical significance. Methods TGF-β1 expression was analyzed by quantitative realtime PCR (qRT-PCR) and Western blotting in tissue samples from 12 cases of LST, 12 cases of adenomal polypus and 16 surgical cases of coloreetal cancer. The relationship between Smo expression and clinicopathological features was in- vestigated. Results The results of qRT-PCR suggested that mRNA expression of TGF-β1 in LST was significantly higher than that in adenomal polypus, but obviously lower than that in colorectal cancer. Western blotting showed that TGF-β1 was rarely expressed in normal eolorectal mucosa and adenomal polypus, but significantly highly expressed in eolorectal cancer. Expression of TGF-β1 in LST was higher than that in normal eolorectal mucosa and adenomal polypus, but lower than that in colorectal cancer. Conclusion TGF-β1 was higher in LST than that in colorectal mucosa and adenomal polypus, meanwhile, it was absolutely different with the high expression of TGF-β1 in colorectal cancer. So the TGF-β1 expression may serve as an important indicator for the malignant trend of LST, and guide follow up proposals.
