摘要
目的探讨前列地尔 (PGE1)对梗塞心肌的保护作用。方法 2 4只麻醉兔随机分为四组 :(1)模型组 ;(2 )PGE12 .5 μg/kg组 ;(3)PGE15 μg/kg组 ;(4 )心得安组。观察结扎左冠状动脉前降支(LAD)后各组内皮素 (ET)、一氧化氮 (NO)、心电图的动态改变 ,同时观察用药各组梗塞面积、心肌病理的改变。结果 (1)结扎LAD后 ,模型组动物ET浓度于 2 4小时明显升高 ,NO则明显降低 ,72小时后均仍未恢复至术前水平。 (2 )静注PGE15 μg/kg后 ,血浆ET浓度明显降低 ,NO浓度明显升高 ,心得安则对ET、NO浓度无明显影响。 (3)PGE15 μg/kg以及心得安使抬高的ST段更快地恢复到等电位线水平且明显缩小心梗范围。结论心肌梗塞时 ,内皮细胞功能失调 ,PGE1能改善内皮细胞内分泌功能 ,缩小心梗范围且不影响心率 ,心得安虽能改善心肌缺血 ,但不能改善内皮细胞内分泌功能。
Objects [WT5”BZ]To investigate the therapeutic effect of prostaglandin E 1(PGE 1) and propranolol on ischemic myocardium in animals. [WT5”HZ]Methods [WT5”BZ]24 rabbits were divided into 4 groups ( n =6 ): 1. Control group. 2. PGE 1 2.5μg/kg group. 3. PGE 1 5μg/kg. qroup 4. Propranolol group. Plasma ET and NO were measured in 24, 72 hours after blocking the flow of coronary artery in each group. ECG of each group was recorded at different time. 72 hours later, the infarct size of myocardium was measured and pathologic change was observed. [WT5”HZ]Results [WT5”BZ]After blocking the flow of coronary artery, the concentration of plasma ET increased and NO decreased significantly in control group. ST segments elevated immediately in all rabbits of blocking coronary artery. PGE 1 (5μg/kg) and propranolol declined ∑ST and diminished infarct size. After injecting PGE 1 (5μg/kg),the concentration of ET decreased,meanwhile the conentration of NO in plasma increased. However, propranolol had no effect on ET and NO. [WT5”HZ]Conclusion [WT5”BZ]PGE 1 could not only alleviate myocardium ischemia, but also recover endothelium function. Propranolol reduced infarct size without affecting the secretion of ET and NO. [WT5”HZ]
