基因重组丝虫融合蛋白抗原检测晚期丝虫病血清特异性抗体的实验研究

摘要 目的评价基因重组丝虫融合蛋白抗原用于检测晚期丝虫病血清特异抗体的价值。方法以基因重组丝虫融合蛋白抗原Chitinase,SXP1,ELISA和westernblotting方法检测 80例晚期丝虫病者血清特异性抗体。结果Chitinase ,SXP1检测晚期丝虫病者血清特异性抗体的阳性率为 2 5~ 40 % ,Mf xt则为 10~ 2 5 % ,敏感性明显高于Mf xt(P <0 0 1)。SXP1可检测到病程为 30年的晚期丝虫病者的血清抗体 ,Chitinase可检测到 10~ 2 0年病程的血清抗体 ,Mf xt仅可测到病程为 10年内的血清抗体。westernblotting可识别出象皮腿血清特异抗体的蛋白分子为 2 1KDa,乳糜尿为 10 0KDa和 95KDa ,鞘膜积液为 11KDa和 6KDa。结论基因重组丝虫融合蛋白抗原可用于晚期丝虫病血清特异性抗体的检测。 Objective To evaluate immunological responses and useful value of chronic cases of lymphatic filariasis (CCLF) through to measure serum specific antifilarial antibodies with recombinant Chitinase and SXP 1 fusion protein antigens. Methods 120 sera samples from CCLF (chyluria, hydrocoele, elephantiasis) and control were measured by ELISA with recombinant antigens and western blotting.Results The positive rates of CCLF serum specific antifilarial antibodies with Chitinase, SXP1 and Mf-xt antigen were 25~40% and 10~25%. The results showed that the sensitive of recombinant antigens was higher than Mf-xt to measure CCLFserum antifilarial antibodies. The course of disease of CCLF for 30 years, 10~20 years and 10 years of serum antifilarial antibodies were detected by SXP 1, Chitinase and Mf-xt respectively. The elephantiasis 21Kda, chyluria 100Kda, 95Kda, and hydrocoele 11Kda, 6Kda were measured by western blotting.Conclusion The recombinant fusion protein antigens can be used for detection specific antifilarial antibodies of CCLF.
