摘要
目的 探讨体外循环期间抑肽酶对血小板保护作用的分子学机制。方法 2 0例健康供血者的静脉血注入含枸橼酸或肝素的试管中 ,用流式细胞术检测抑肽酶对血小板膜糖蛋白GPⅡb Ⅲa受体及P 选择素表达的影响。结果 抑肽酶在 5 0~ 2 0 0U/ml时可减少ADP和凝血酶受体激活肽 6激活的血小板膜糖蛋白GPⅡb Ⅲa受体的表达 (P <0 0 5 ) ,但对P 选择素表达无影响。结论 抑肽酶减少激动剂诱导的活化血小板膜糖蛋白GPⅡb Ⅲa受体的表达 ,而GPⅡb Ⅲa受体在血小板聚集及其粘附于生物材料表面的过程中起重要作用。
Objective To invistigate molecular mechanism of the effect of aprotinin on platelets membrane glycoproyin during cardiopulmenary bypass Methods The venous blood of 20 healthy volunteers in the tubes containing 3 2% trisodium citrate or 5u/ml heparin,to evaluate the effect of aprotinin on the expressions of platelets membrane glycoprotein (GP) Ⅱb Ⅲa receptor and P seclectin with flow cytometric analysis Results Aprotinin at 50 200U/ml decreased the expression of activated GP Ⅱb Ⅲa complex in response to adenosine diphosphate or thrombin receptor activator peptide 6 in a dose dependent manner in both citrated and heparinized whole blood experiments (P<0 05) There was not influence of aprotinin on the expression of P seclectin Conclusion The aprotinin decreases the agonist induced expression of activated GP Ⅱb Ⅲa receptor that play a major role in platelet aggregation and adhesion to biomaterial surfaces
