摘要
目的探讨IL-10对A7r5大鼠胸大动脉平滑肌细胞增殖的抑制作用及其与p53、bcl-2的变化关系。方法用MTT法测定细胞抑制率;倒置显微镜观察细胞形态;细胞免疫组化法和Western bolt观察细胞内p53、bcl-2基因的表达。结果 IL-10对AngⅡ诱导的A7r5平滑肌细胞增殖有明显抑制作用,但不呈剂量依赖关系;可逆转p53、bcl-2的表达。结论 IL-10对AngⅡ诱导的A7r5平滑肌细胞增殖有明显防治作用,与细胞凋亡基因p53、bcl-2相关。
Objective To investigate the effect of IL-10 on the proliferation of vascular smooth muscle A7r5 cells induced by AngⅡ and the expression of p53 and bcl-2.Methods The cell activity was assessed using MTT method.The cellular morphology was observed by inverted phase-contrast microscope.The expression of p53 and bcl-2 was detected by immunohistochemical method and western blot.Results IL-10 significantly inhibited A7r5 cell proliferation,but not in a dose-dependent manner,and reversed both p53 and bcl-2 expression.Conclusion IL-10 can prevent the vascular smooth muscle A7r5 cell proliferation induced by AngⅡ,which is related to modulation of the expression of p53 and bcl-2.
