高效液相色谱法测定肌苷片的溶出度

Determination of Dissolution of Inosine Tablets by HPLC

目的建立肌苷片溶出度的高效液相色谱（HPLC）测定方法。方法色谱柱为Agilent,clipse XDB-C18（4.6mm×250mm,5μm）;流动相：甲醇-水（10：90）;流速：1.0mL/min;检测波长为248nm;柱温为25℃。结果肌苷片在2.54~40.60μg/mL的浓度范围内呈良好线性关系（r=0.999 8）,平均回收率为100.1%（n=9）,RSD为0.3%。结论该方法准确、可靠,与标准方法比较,结果较为满意,可用于肌苷片的溶出度测定。

Objective To establish an HPLC method for dissolution of inosine tablets. Methods Agilent C18 column（4.6 mm×250mm, 5μm）was used with a mobile phase composed of methanol-water（10： 90） ）at the flow rate of 1.0 mL,min-l.The detection wavelength was 248 nm and the temperature of the column was 25 ℃ .Results The calibration curve of inosine was linear in the range of 2.54-40.60μg·L^-1（r=0.999 8） ）.The average recovery was 100.1% with an RSD of 0.3% （n=9）. Conclusion Compared with the standard method, the results were satisfactory, and the method was accurate and reliable, which could be used for the determination of dissolution of inosine tablets.
