摘要
目的 :观察中药海参猴桃液(SSAP)辅以少量rIL-2激活的LAK细胞回输家兔后 ,其T细胞亚群水平情况 ,为从理论和实践探讨中药肿瘤免疫的作用及其原理提供依据。方法 :采取健康家兔下肢静脉血分离淋巴细胞作待培养细胞 ,分设实验组(中药SSAP3000ug/ml+rIL-2100u/ml)、阳性对照组(rIL -21000u/ml)、空白对照组(加等量生理盐水) ,培养第6天起回收LAK细胞分别回输给3组家免 ,回输6次后 ,采取家兔外周静脉血 ,送化验检测T细胞亚群 ,观察各组在回输LAK细胞前后CD3、CD4、CD8、CD4/CD8 水平情况。结果 :在回输后各项指标中 ,实验组及阳性对照组与空白组比较 :实验组及阳性对照组的CD3、CD4、CD4/CD8 各项指标高于空白组 ,统计学P<0 05 ,差异有显著性 ;实验组及阳性对照组的CD8 值高于空白组 ,但P>0 05 ,差异无显著意义 ;实验组与阳性对照组的CD3、CD4、CD8、CD4/CD8 水平相当 ,统计学P>0 05,差异无显著性。结论 :中药海参猴桃液(SSAP)辅以少量rIL -2激活的LAK细胞与单独rIL-2激活的LAK细胞回输家兔后 ,对T细胞亚群的调节具有相似的影响作用。说明SSAP液能协同少量rIL-2刺激机体免疫系统 ,增强机体的细胞免疫功能 ,提高机体的免疫力。
Purpose: To observe the T-cell subpopulations level after Hai Shen Hou Tao Ye (SSAP) plus a small dosage of LAK cells activated by rIL-2 was infused back into rabbits so as to examine their effects on tumor immunity and study their mechanism. Methods: Lymphocyte separated from venous blood of healthy rabbits was used as cultural cells. Three groups were formed—— the experiment group with SSAP 3 000ug /ml+rIL-2 100u/ml, the positive control group with rIL-2 100u/ml and the control group with equal amount of normal saline. The LAK cells were infused back into the rabbits of these 3 groups after 6-day culture. After 6 times of infusion were given, the peripheral venous blood was taken to examine T-cell subpopulations and observe the changes in the levels of CD3 、CD4 、CD8 and CD4/CD8 before and after the infusion. Results: After the infusion, the indexes of CD3 、 CD4 and CD4/CD8 in the experiment group and the positive control group were higher than those in the control group with the significant difference(P<0.05).Also the level of CD8 in the experiment group and the positive control group was higher than that in the control group, but no significant difference was indicated (P>0.05).The levels of CD3 、 CD4 、CD8 and CD4/CD8 both in the experiment group and the positive control group were quite close with no significant difference(P>0.05). Conclusions: LAK cells activated by SSAP with a little rIL-2 and LAK cells activated only by rIL-2 have similar effects in regulating T-cell subpopulations after they are infused back to rabbits. It suggests that SSAP with a little rIL-2 can stimulate the immune system, improve the cell's immunologic functions and immunity of the organ.
