基因检测大肠癌区域淋巴结微转移的研究 被引量:5

Genetic detection of regional lymph node micrometastases of colorectal carcinoma.

摘要 目的 探讨大肠癌区域淋巴结微转移的检测和临床意义。方法 采用逆转录聚合酶链反应 (RT -PCR)技术 ,扩增角蛋白 19(K19) ,对 12例病人大肠癌肿瘤组织和 95个区域淋巴结进行检测。结果  12例病人大肠癌组织均有K19mRNA表达 ,良性病人的 2 1个淋巴结均无表达。大肠癌区域淋巴结 11个经病理检查有转移 ,其K19mRNA表达阳性。 84个病理检查未发现转移的淋巴结中有 8个K19mRNA表达阳性。结论 K19mRNART -PCR方法对于大肠癌区域淋巴结微转移灶有辅助诊断价值。 Objective To discuss the detection of tumor micrometastases in regional lymph node of colorectal carcinoma and its clinical significance.Methods The expression of keratin 19(K19) mRNA was measured in tumor tissues of 12 cases and 95 regional lymph nodes from colorectal carcinoma patients by using reverse transcriptase-polymerase chain reaction(RT-PCR).Results K19 mRNA expression was detected in all 12 colorectal carcinoma,but not in 21 benign lymph nodes.11 lymph nodes were found to have metastases by histological staining and K19 RT-PCR.Of the remaining 84 negative lymph nodes,8 lymph nodes were with positive expression of K19 mRNA.Conclusion The K19 mRNA RT-PCR method can sensitively detect regional lymph nodes micrometastases and is helpful to the diagnosis.
