摘要
目的研究大鼠肝硬化形成过程中细胞色素P450酶(cytochrome P450,CYP450)基因的mRNA表达水平变化。方法采用四氯化碳(CCl_4)腹腔注射建立肝硬化大鼠模型,设置空白对照组、溶剂对照组、CCl_4造模组,定期采集各组大鼠肝脏组织样本,通过荧光定量PCR的方法测量大鼠肝脏组织中目标基因相对的mRNA水平。结果 CCl_4造模组肝脏组织的CYP2c23、CYP2d6和CYP3a23的mRNA表达水平较空白对照组和溶剂对照组均显著升高(P<0.01)。这些基因的mRNA表达水平与肝脏受到CCl_4损伤的周期长短无显著相关性。结论肝脏组织中CYP2c23、CYP2d6和CYP3a23基因在肝硬化模型中的表达水平显著高于正常肝脏组织中的水平。
Objective To study the gene expression patterns of cytochrome P450 (CYP450) genes,including CYP2c23, CYP2d6 and CYP3a23, in acute CC14 liver damage model. Methods Untreated group, olive feeding group and CC14 treated group were established, mRNA levels of targeted genes were relatively quantitated by applying quantitative reahime(qPCR) PCR method. The quantitative PCR data were used to evaluate the effect of CC14 on the gene expression of the target genes. Results The gene expressions of CYP2c23, CYP2d6 and CYP3a23 in CC14 treated group were significantly increased compared to those in untreated group and olive feeding group ( P 〈 0.01 ). And the effect of CC14 on gene expression was not time-dependent. Conclusion The gene expression levels of CYP2c23, CYP2d6 and CYP3a23 in liver can be elevated in acute CC14 damaging model in SD rats.
