摘要 目的:改进HPLC同时测定柴胡中黄酮和皂苷含量的方法。方法:采用MERCK PUROSPHER STARC18色谱柱(250mm×4.6mm,5μm);流动相为甲醇-0.05%磷酸(梯度洗脱);流速为1.0mL·min-1;柱温为35℃;检测波长为210nm。结果:2个黄酮成分(芦丁、槲皮素)和2个皂苷成分(柴胡皂苷a、柴胡皂苷d)分别在0.36—9.09μg(r=0.9997)、0.47—11.70μg(r=0.9995)、1.15—28.66μg(r=0.9998)和0.66~16.46μg(r=0.9998)线性关系良好;平均回收率为96.39%,97.44%,100.04%,97.89%,RSD均小于1.0%。结论:本方法简便准确、稳定可靠,可以用于同时测定柴胡中黄酮和皂苷成分的含量。 Objective: To detect flavonoids and saponins content in Bupleuri Radix by improved HPLC method. Methods: Using MERCK PUROSPHER STAR C18 column (250 mm ×4.6 mm, 5 μm) ; the mobile phase was methanol-0. 05% phosphate (gradient elution) ; flow rate was 1 mL.min-1 ; column temperature was 35 ℃ ; the detection wavelength was 210nm. Results: 2 flavonoids (rutin, quercetin ) and 2 saponins (saikosaponin a, saikosaponin d) in 0.36-9.09 μg(r =0.999 7), 0.47-11.70 μg (r =0.999 5), 1.15-28.66 μg(r =0.999 8) and 0. 66-16. 46 μg(r =0. 999 8) linear ranges; the average recovery rate was 96.39%, 97.44% , 100. 04% and 97. 89% respectively, all RSD 〈 1.0%. Conclusion: This method is simple, accurate, stable and reliable, Can be used for the simultaneous determination of flavonoids and saponins in Radix Bupleuri.
