肺癌Th1/Th2免疫反应状态的研究

Study of reaction state of Th1/Th2 of patients with lung cancer

为探讨肺癌细胞及肺癌患者外周血单个核细胞 (PBMC) Th1/ Th2的免疫反应状态 ,为肿瘤的免疫治疗提供依据 ,以 IL - 2和 IFN- r代表 Th1型细胞因子 ,IL - 4、IL - 6和 IL - 10代表 Th2型细胞因子 ,用 RT- PCR方法检测了三种人肺癌 (鳞癌、腺癌、小细胞癌 )细胞株及 2 3例肺癌患者 PBMC中 Th1/ Th2型细胞因子 m- RNA的表达。结果显示 ,三种肺癌细胞的 Th1型因子 IFN- r和 IL- 2均无表达 ,而 Th2型因子 IL- 4、IL- 6、IL- 10均表达 ;肺癌患者PBMC中 IL- 4、IL- 6和 IL- 10表达阳性率均较正常对照组显著增高 ,P均 <0 .0 5。而 IL- 2无 1例表达 ,IFN- r仅 1例表达。证实肺癌细胞株及肺癌患者 PBMC均呈现

The reaction state of Th1/Th2 in human lung cancer cells and peripheral blood mononuclear cells(PBMC)from lung cancer patients was studied for immunological treatment of cancer the m RNA expression of type Th1 cytokines(IL 2,IFN r)and type Th2 cytokines(IL 4,IL 6,IL 10)was detected by reverse transcriptase PCR in the lung cancer cell strains(including sq uamate and adenogenous and small cell lung cancer)and the PBMC from 23 lung cancer patients and 19 healthy controls The result showed that none of the human lung cancer cell strains expresses m RNA of IL 2 and IFN r,whereas all type Th2 cytokines(IL 4,IL 6,IL 10)were presented The m RNA expressing rate of IL 4,IL 6,IL 10 of PBMC from lung cancer patients was obviously higher than that from healthy control,P<0 05 respectively No m Rna of IL 2 was expressed in patients's PBMC,and a positive expression of IFN r was found in only one patient's PBMC It is suggests that there is a predominant reaction of type Th2 in both lung cancer cells and PBMC from lung cancer patients
