摘要
目的 :观察急性重症胆管炎 (ACST)大鼠肺组织内一氧化氮合酶 (NOS)、肺泡巨噬细胞分泌一氧化氮 (NO)的变化及活血清解灵的调节作用。 方法 :通过胆总管远端结扎 ,近端注入菌液并封闭的方法 ,造成大鼠ACST模型。采用单纯胆管减压、胆管减压配合活血清解灵两种不同方法治疗。分离和培养肺泡巨噬细胞 ,采用Griess法检测收集培养液中NO水平。应用分光光度法测定NOS活性。 结果 :ACST组肺内NOS的水平和肺泡巨噬细胞分泌NO的水平显著高于假手术对照组 (P <0 .0 5 ) ,中药活血清解灵治疗组亦高于假手术对照组 ,但明显低于ACST组 (P <0 .0 5 ) ,减压组与ACST组无差异 (P >0 .0 5 )。 结论 :活血清解灵具有调节NOS的作用 ,抑制肺泡巨噬细胞过度释放NO ,减少NO对组织细胞的损伤。
Objective:To observe the adjustment of “Huo Xue Qing Jie Ling”on the changes of NO secreted by the pulmonary alveoli macrophage and NOS in the lungs of acute cholangitis of severe type(ACST)model rat.Methods:ACST model was made by distally ligating the choledochus and proximally injecting bacteria suspension.After the two kinds of therapy,including drainage of the common bile duct(CBD)and drainage of the CBD with administration of Chinese herbal compound formula:“Huo Xue Qing Jie Ling”,the level of NO secreted by the pulmonary alveoli macrophage and NOS in the lungs was observed.Results:The level of NO secreted by the pulmonary alveoli macrophage and the level of NOS in the lungs in the ACST model group was significantly higer than that in the pseudo operation contrast group(P<0.05).The above substance in the Chinese herbal group was higher than that in the pseudo operatioin group,but lower than that in the ACST model group(P<0.05).The difference between the common bile duct group and the ACST model group was not remarkable(P>0.05).Conclusion:“Huo Xue Qing Jie Ling”could modulate NOS,inhibit the pulmonary alveoli macrophage to excessively release NO and alleviate the tissue and cell lesion caused by NO.
