应用ClinProt系统分析结肠腺癌患者血清蛋白表达谱 被引量:1

摘要 目的寻找可能与结肠腺癌相关的血清候选蛋白,为结肠腺癌的综合防治提供重要的理论基础和手段。方法收集36例结肠腺癌患者和22例正常对照组血清,经Clinprot磁珠纯化,基质辅助激光解析电离飞行时间质谱(MALDI-FOF-MS)分析建立相应的蛋白质指纹图谱,应用Clinpro Tools生物信息学方法研究其血清蛋白表达谱。对不同质荷比(M/Z)的差异蛋白通过在数据库中的分析比对,筛选出可能与结肠腺癌发病相关的血清候选蛋白。结果结肠腺癌和正常对照组血清样本蛋白丰度平均变异系数为0.229,与对照组比较,结肠腺癌组有15种蛋白上调,7种蛋白下调。选取差异最为明显的两个蛋白:M2369.93Da和M6051.12,建立结肠腺癌组和正常对照组差异表达蛋白平均谱图,采用GA模型算法,选取差异蛋白,结肠腺癌阳性和阴性识别率为79.7%,准确性结肠癌阳性组为96.1%,结肠癌阴性组为85.5%,预测能力为72.8%。结论通过液体蛋白芯片飞行时间质谱系统作为分析工具,发现15种蛋白上调,7种蛋白下调,这些蛋白可能为潜在的结肠腺癌肿瘤标志物。 Objective To find out the serum proteins related to the onset of adenocarcinoma of colon which provide important information for integrally mitigating adenocarcinoma of colon. Methods We collected blood serum from 38 cases of preoperative adenocarcinoma of colon patients and from 22 normal controls, using Clinprot magnetic bead purification, matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF-MS) to analysis corresponding proteins. We analyzed serum protein spectrum by ClinProTools bioinformatic method. Results The mean coefficients of variance (CV)for protein peak intensities in esophageal cancer group and normal control were 0.229. Compared with control group, adenocarcinoma of colon group had 15 up-regulated and 7 down- up-regulated. At last, mass charge ratio (M/Z) 2369.93 and 6051.12 was screened as category variable to establish category predictive model. Conclusion Magnetic bead based sample fractionation and MALDI-TOF-MS is a tool to research protein expression map and has very high repeatability and precision. The expression of 15 proteins is up-regulated in adenocarcinoma of colonr. The expression of 7 proteins is down-regulated in adenocarcinoma of colon. These proteins maybe potential tumor markers of adenocarcinoma of colon.
