摘要
应用免疫组化和同位素点杂交技术对 2 5例甲状腺乳头状癌新鲜冰冻组织及 19例其他类型甲状腺肿瘤和非肿瘤组织中 Neu癌基因 p185蛋白和 m RNA的表达进行研究。乳头状癌中 17例 p185癌蛋白表达阳性 ,而其他类型肿瘤和非肿瘤组织均未见 p185表达 ;乳头状癌和淋巴结转移灶中 Neu癌基因 m RNA的表达水平明显高于其他肿瘤和非肿瘤组织。而且石蜡包埋的乳头状癌标本中 Neu癌基因蛋白表达率明显低于新鲜标本。认为 Neu癌基因 p185蛋白的表达是甲状腺乳头状癌的一个特征性改变 ,Neu蛋白的表达与 m RNA的表达相一致 ,Neu癌蛋白检测的稳定性取决于组织标本的质量。
WT5”BZ]To investigate the expression and relationship of p185 oncoprotein and mRNA of Neu oncogene in human thyroid carcinomas,p185 oncoprotein and mRNA of Neu oncogene were detected in 25 cases of thyroid papillary cacinoma and 19 cases of the other thyroid tumors and non neoplastic specimens on frozen tissue using immunohistochemistry and isotope Dot blot hybridisation p185 immunoreactivity was detected in 17 out of 25 papillary carcinomas,while no Neu oncoprotein immunstaining was seen in cases of the other thyroid tumors and non neoplastic tissue Papillary carcinomas and lymph node metastases showed significantly higher levels of Neu mRNA compared to the other thyroid tumors and non neoplastic tissue The positive rate of Neu oncoprotein on formalin fixed,paraffin embedded material were much lower than levels on frozen tissue This suggests that p185 oncoprotein expression of Neu oncogene may be a feature of papillary thyroid carcinomas,and is consistent with mRNA expression The detecting stability of Neu oncoprotein is depend on optimal tissue quality [WT5”HZ]
