摘要
目的 观察小鼠胸腺树突状细胞 (TDC)的形态。 方法 密度梯度离心结合小鼠 CD5单抗淘洗法(panning) ,分离出小鼠 TDC并行光镜及电镜观察。 结果 台盼蓝排斥试验示 90 %以上细胞存活。光镜下发现初分离的 TDC常和周围的胸腺细胞形成紧密的细胞簇。S- 10 0蛋白染色见 TDC胞体和突起内有棕黄色反应产物。ANAE反应呈阴性或在核附近有数个小颗粒反应。ATP酶反应阴性。电镜见细胞表面有许多树状突起 ,核形不规则 ,胞质电子密度低 ,有丰富的线粒体、核糖体及完整的高尔基复合体。 结论 所分离的
Objective\ To observe the morphology of mouse thymic dendritic cells(TDC). Methods\ TDCs were isolated from thymic suspension by Ficoll\|Hypaque gradients and Percoll gradients of different densities followed by Panning procedure with anti\|CD5 monoclonal antibodies. The cells in the TDC\|enriched fraction were assayed. Results\ There were more than 90% of surviving cells in this fraction. It was found that TDC could contact and cluster with thymocytes. S\|100 reaction product was detected primarily in the cytoplasm and cytoplasmic processes of these cells. In the ANAE reaction, some cells were negative, and some contained small cluster of positive tiny granules located closely to the nucleus. The ATPase activity was negative. Most cells exhibited many bulbous protrusions. The nucleus was irregular in shape. The cytoplasm was often electron\|lucent, but contained large amount of free ribosomes, mitochondria and well\|developed Golgi complexes. Conclusion\ The obtained TDCs had the classical morphology of dendritic cells.\;
