摘要
利用国内外猪精液常温保存的研究成果 ,设计了 4个公猪精液稀释液配方(T1、T2、T3、T4) ,以目前四川农村猪人工授精中常用的EDTA液配方为对照 ,在室温条件下 (1 6~ 2 8℃ )通过逐日镜检的方法检查精子活力 ,每个配方 5次重复 ,筛选出了保存期达 5天、精子活力 0 5级以上的配方 2个 (T2、T3) ;保存期达 6天、精子活力 0 5级以上的配方 1个 (T4)。使用T4配方保存 6天的稀释精液输精母猪 6 8头 ,经超声波妊娠诊断仪检测母猪受胎率达 86 8%。对 4个精液稀释液配方在溶解后90min内的pH值变化进行了研究 ,各稀释液在溶解后的 6 0min内 pH值出现明显波动 ,之后达到平衡 ,表明稀释液宜在使用前
Four boar semen extenders were designed(T1,T2,T3,T4)to select long time boar semen extender using EDTA extender as the control,which was quite popular in swine artificial insemination in Sichuan Province.By means of inspecting sperm activity with microscope day in day at room temperature(16~28℃),two extenders(T2,T3)were confirmed to be able to preserve sperm activity above 0 5 for five days,and one extender(T4)to be able to preserve sperm activity above 0 5 for six days.In order to check its preserving effect,sixty eight sows/gilts were mated with diluted semen preserved by T4 for six days.The pregnant rate was 86 8%.In order to better guide the usage of extender more effectively,the changes in pH of the four extenders(still using EDTA as the control)were measured during the first 90 minutes after preparation.Though boar semen extenders do not have the same pattern of pH change across the time,they took almost the same time(60 minutes)to reach balanced pH.Thus it was suggested that the semen should not be mixed with extender until a stable pH was reached.In general,a liquid boar smen extender should be prepared 60 minutes prior to mixing with raw semen.
