摘要
利用 Qiagen微量植物 DNA提取试剂盒 ,在 1小时内即可从植物组织获得总 DNA。提取过程中勿需酚 /氯仿和 SDS抽提 ,操作简便、快捷。所得 DNA样品的 OD2 60 /OD2 80 值在 1 .7— 1 .9之间 ,样品纯度高 ;该样品不含 PCR反应抑制剂及其他酶反应抑制剂 ,可被各种限制性内切酶完全降解 ,适合于 PCR、印迹、RAPD、AFLP和 RFLP分析等各种下游应用。
A fast and simple way to isolate total DNA from up to 100mg of plant tissue was developed in the present paper. By using the DNeasy Plant Mini Kit, high quality of total DNA from plant was yielded in less than 1 hour. Purification required no phenol or chloroform extraction and involved minimal handling. This made the kit ideal for simultaneous processing of multiple samples. Plant DNA isolated with this method could be completely digested by restriction endonucleases, and was ready for use in all kinds of downstream applications including PCR, blotting, analysis of RAPD, AFLP and RFLP.
