补肾活血法与益气补血法对骨髓抑制小鼠造血细胞作用的比较

A comparative study of Bushen Huoxue recipe and Yiqi Buxue recipe on hematopoiesis regulation of myelosuppression in mice

目的比较补肾活血法与益气活血法对化疗后骨髓抑制模型小鼠造血细胞作用特点与差异。方法将SPF级ICR小鼠随机分为正常对照组、模型组、补肾活血组和益气补血组,灌胃1次/d,连续给药至实验结束,第8天采用腹腔注入环磷酰胺(100mg/(kg·d)),连续3d造模。造模后第1天和第7天各处死半数动物,检测骨髓有核细胞计数、骨髓CD_(34)^+细胞及细胞周期。结果两中药组均可提高骨髓有核细胞数,增加骨髓细胞增殖指数;其中益气补血法对造血细胞动员能力更强,补肾活血法对维持骨髓造血细胞增殖效果更佳。结论补肾活血法与益气活血法均可以促进骨髓抑制小鼠造血功能恢复,其作用机制存在差异。

Objective It is to investigate the curative effects and features between Bushen Huoxue recipe and Yiqi Buxue recipe on hematopoiesis regulation of bone marrow suppression induced by eyclophosphamide （CTX） in mice. Methods SPF grade ICR mice were used and randomly divided into 4 groups： normal control group, model group, Bushen Huoxue group, Yiqi Buxue group. The mice were chronically administrated normal saline, or Bushen Huoxue Recipe, or Yiqi Buxue Recipe once a day continually till the experiment ended. The experimental models of myelosuppression were established at day 8 by ip cyclophosphamide （CTX） 100 mg/kg for 3 days continuously. BMNC, CD34 ＋ cells and cell cycle percentage were detected on the first day and 7th days after the successful model set. Results Both of the two Chinese medicine treatment can significantly elevating the hematopoietic stem cells number and promoting the proliferation of bone marrow cells in myelosuppression mice; Yiqi Buxue recipe was better at promoting the polfieration of hematopoietie stem, and Bushen Huoxue recipe was more effective in regulating cell cycle. Conclusion Bushen Huoxue recipe and Yiqi Buxue recipe can both promote the recovery of hematopoi- etic function in myelosuppression mice, but their action mechanisms are different.
