摘要
目的 探讨骨碎补总黄酮对聚乙烯微粒引起单核细胞分泌肿瘤坏死因子-α(TNF-α)的影响.方法 采集20例健康人体外周血,分离单核细胞,随机分组培养.A组(对照组):仅单核细胞;B组:单核细胞+聚乙烯微粒;C组:单核细胞+聚乙烯微粒+帕米磷酸钠(10 mg/L);D组:单核细胞+聚乙烯微粒+骨碎补总黄酮(5 mg/L);E组:单核细胞+聚乙烯微粒+骨碎补总黄酮(10 mg/L);F组:单核细胞+聚乙烯微粒+骨碎补总黄酮(20 mg/L).培养48 h后,检测细胞上清液中TNF-α的含量.结果 TNF-α含量B组高于A组(P〈0.01);C、D、E、F组分别低于B组(P〈0.01);C组与D、E、F组之间的差异无统计学意义(P〉0.05);D组和E组之间的差异无统计学意义(P〉0.05),E组和F组之间的差异无统计学意义(P〉0.05),F组低于D组(P〈0.01),随着药物浓度的增加,D、E、F组有逐渐减少趋势.结论在体外聚乙烯微粒可以刺激人外周血单核细胞过量分泌TNF-α.在体外骨碎补总黄酮和帕米磷酸钠均可有效抑制人外周血单核细胞过量分泌TNF-α.在一定剂量范围内,在体外骨碎补总黄酮和帕米磷酸钠抑制人外周血单核细胞过量分泌TNF-α的效果相似.骨碎补总黄酮能有效抑制由磨损微粒介导的TNF-α的高表达,有望成为将来防治人工关节无菌性松动的一种很有潜力的药物.
Objective To explore the possibility of using osteopraetic total flavone (OTF) to prevent and treat asep-tic loosening of prothesis. Methods Blood samples obtained from 20 healthy human donors were processed to obtain the monocytes. The monocytes were divided into 6 groups. Group A: only monocytes; Group B: monoeytes + polyethylene particles; Group C: monocytes + polyethylene particles + pamidronate ( 10 mg/L) ; Group D: monocytes + polyethylene particles + OTF (5 mg/L) ; Group E: monocytes + polyethylene particles + OTF (10 mg/L) ; Group F: monocytes + po-lyethylene particles + OTF (20 mg/L). The monocytes in each group were cultured for 48 h and the levels of TNF -α in each group were determined. Results The levels of TNF - α were much higher in group B than in groups A, C, D, E, and F ( P 〈 0.01 ). There was no significant difference between group C and groups D, E, and F ( P 〉 0.05 ). There was no significant difference between group D and group E (P 〉 0.05), also there was no significant difference between group E and group F (P 〉 0.05). The levels of TNF - α were much lower in group F than in group D (P 〈 0.01 ). With the increasing dose of OTF, the levels of TNF - α were decreased by OTF gradually in groups D, E, and F. Conclusion OTF can effectively inhibit TNF - α secretion of the moncytes stimulated by polyethylene particles in vitro. In a certain dose range, OTF which is similar to pamidronate can inhibit the secretion of TNF - α in vitro. OTF appears to be a poten-tial therapeutic candidate for the treatment and prevention of aseptic loosening of prothesis.
