3'-大豆苷元磺酸钠对去卵巢大鼠骨代谢的影响 被引量:3

摘要 目的观察3'-大豆苷元磺酸钠对去卵巢大鼠骨代谢的影响。方法选择健康SD雌性大鼠60只,随机分成6组,每组10只:①假手术组;②卵巢切除组(模型组);③卵巢切除加尼尔雌醇组(阳性对照组);④卵巢切除加3'-大豆苷元磺酸钠低剂量组(25μg·kg-1);⑤卵巢切除加3'-大豆苷元磺酸钠中剂量组(50μg·kg-1);⑥卵巢切除加3'-大豆苷元磺酸钠高剂量组(100μg·kg-1)。连续给药6周,末次给药24 h后实验,麻醉后取血清,测定血清中骨钙素、碱性磷酸酶、抗酒石酸酸性磷酸酶和孕酮的含量。结果模型组和3'-大豆苷元磺酸钠高剂量组骨钙素分别为(5.04±0.05)和(4.82±0.05)ng·mL-1;碱性磷酸酶分别为(47.31±0.02)×10-2和(36.90±0.21)×10-2U·mL-1;抗酒石酸酸性磷酸酶分别为(1.72±0.11)和(3.02±0.13)U·L-1;孕酮分别为(49.72±2.00)和(53.41±1.12)ng·mL-1。3'-大豆苷元磺酸钠可明显降低血清中骨钙素及碱性磷酸酶的含量(P<0.01),可明显升高血清抗酒石酸酸性磷酸酶(P<0.05)和血清孕酮含量(P<0.01)。结论 3'-大豆苷元磺酸钠可促进骨形成,从而对骨代谢发挥调节作用。 Objective To investigate the effects of 3'-daidzein sulfonate sodium on bone metabolism of ovariectomized rats. Methods Sixty female SD rats were selected and randomly divided into six groups:sham-operation (normal control), ovariectomized model (model), ovariectomized with nilestriol (positive control ), high-dose, middle-dose, and low-dose ( ovariectomized with 3'-daidzein sulfonate sodium 100, 50 and 25 μg · kg-1 , respectively). After continuous administration for 6 weeks and 24 h after the last administration, the serum was obtained after anesthesia and contents of the osteocalcin ( BGP), alkaline phosphatase(AKP), tartrate-resistant acid phosphatase (STrACP), and progesterone were determined. Results In the model control and high dose groups, respectively, the BGP concentration was (5.04±0.05) and (4.82±0.05) ng·mL-l, AKP was (47.31±0.02) ×l0-2and (36.90±0.21) ×10-2 U· mL-1, STrACP was (1.72±0.11) and (3.02±0.13) U. L-l, and progesterone was (49.72±2.00) and (53.41 ± 1.12) ng · mL-1. 3 ’-daidzein su/fonate sodium decreased the content of serum osteoealcin and AKP significantly ( P〈0.01 ), but elevated STrACP ( P〈0.05 ) markedly and increased the content of progesterone ( P〈0.01 ). Conclusion 3'- daidzein sulfonate sodium regulates bone metabolism by promoting bone formation.
